肺癌の抗癌剤耐性獲得機序における組織因子のアポトーシス抑制作用の分子細胞学的研究

研究概要	肺癌患者にはシスプラチンを含む抗癌剤併用療法が施行されているが、多くの場合抗癌剤耐性により早々に抗癌剤が効かなくなり、治療対策の確立のために抗癌剤耐性の分子機構を明らかにすることが必要である。アポトーシスの抑制は癌における抗癌剤耐性の中心的な機構と考えられ、我々の従来の研究においては血液凝固系組織因子(TF)の発現が抗癌剤耐性に関与している可能性を示唆するものである。従来、TFについては外因系凝固の活性化における役割を中心に研究されてきたが、最近になって、TFのアポトーシス抑制作用が注目されてきた。しかし、TFによるアポトーシス誘導と抗癌剤耐性の関係については明らかではない。抗癌剤耐性とTFによるアポトーシスの関連を明らかにするために我々は平成13年度に先ず野生型TF(wild-TF)cDNA及び自己リン酸化を阻害する細胞内ドメインの変異型TF(mutant-TF)cDNA高発現ベクター(pCR3.1)をTAクロニング法で作製した。ベクターのTFcDNAのオリエンテイションとシクエンスを確認した後にそれぞれの発現ベクターを抗癌剤感受性でTF発現の少ないPC9/P肺癌細胞殊にcationic liposome法で導入し、形質変換細胞を調整している。mutant-TF,wild-TFまたはベクターのみのある形質変換細胞を分離している。一方、最近我々は癌患者において血液中の線溶系の阻害剤であるthrombin-activatable fibrinolysis inhibitor(TAFl)はTF活性化の機序に重要な役割果たしていることを報告した。現在、in vitroの実験系で抗癌剤耐性の分子機構におけるTFの関与を検討し、TFとTAFl等の凝固因子との関連も検討している。
