進行動脈硬化症完全退縮を目指した一酸化窒素合成酵素及び関連酵素の複合遺伝子治療

研究概要	eNOS, iNOSの活性制御による進行動脈硬化症の完全退縮を目指して検討を続けた。ENOSでは、細胞内局在(アシル化酵素)と基質、特にBH4に注目して補酵素の供給をみた。律速酵素GTPCH Iの活性及びmRNAが、血管及び培養内皮で測定可能になり、高血糖の影響を培養内皮で検討した。さらに、インスリン、女性ホルモン、スタチン製剤の影響を検討し成果を得た。まあ、アシル化、リン酸化との関連でAKT/PkBとの関連を検討した。活性酸素生成に関与するNADPH oxidase, Xantine/Xanthine oxidase, iNOS, eNOSについて検討した。高血糖下ではeNOSが活性酸素を出す事を明らかにし、上記のGTPCHIIとの関連も明らかになった。動物実験では糖尿病自然発症ラット(OLETF)での実験が進み、腹腔マクロファージを用いて、活性酸素放出能を測定する事が可能になった。OLETFでは、littermateのラットより活性酸素放出能が強く、血管からのNO分泌も低下していた。NO合成酵素阻害剤を同時投与すると、活性酸素放出能が高まり、血管からのNO分泌は殆どなくなった。NADPH oxidase阻害剤を投与するとこれらの反応が元に戻りNO分泌も戻った。NO産生系は活性酸素産生系に変わるし、NADPH oxidaseのような活性酸素産生系自身とも関連しているという新しい成果が理解された。これらの成果は海外共同研究者のIgnarro教授とも討議し賞賛された。彼の研究室でもNO分泌合剤を開発しており共同研究をしている。ERα遺伝子ベクターが完成した。
