高グリシン血症の新しい病因遺伝子の単離

研究概要	高グリシン血症は、グリシン分解の主経路であるグリシン解裂酵素系(GCS)の遺伝学的障害により体液中にグリシンが蓄積し、主に新生児期に重篤な中枢神経障害を引き起こす代謝病の一つである。最近、見出したイスラエル在住の高グリシン血症の近親婚大家系ではGCS構成蛋白をコードする3つの遺伝子(GLDC, AMT, GCSH遺伝子)の全ての蛋白翻訳領域を変異検索したが、アミノ酸の置換を伴う遺伝子変異を見出すことは出来なかった。この家系は新しい責任遺伝子の変異により発症していると考え、責任遺伝子の遺伝子マッピングを開始した。多くのマイクロサテライト・マーカー増幅用の蛍光標識プライマー・セット(Research Genetics社製、平均10cM間隔)にて各家族のDNAを鋳型としてPCR増幅を行う。異なる蛍光物質で標識された3-4個のPCR産物をまとめてAB1373A DNAシークエンサーにて、電気泳動後、GeneScanプログラムにて遺伝子型を決定する。得られた遺伝子型のデータをGene Hunterプログラムにて解析し、lodスコアの算定を行ったが現在のところ高いlodスコアを持つ領域は見出されていない。今後、解析を進め、候補領域と考えられる部分に関しては、更に多くのマイクロサテライト・マーカーによる高密度マッピングを行い、有意に連鎖が認められる染色体領域を同定する予定である。
