肝臓幹細胞を用いたウィルソン病に対する細胞移植治療法の開発

研究概要

1.目的
(1)申請者らが確立した方法によって分離した、正常な機能を保持していると考えられるラット肝臓幹細胞(肝上皮細胞)を移植に適した状態に加工し、充分な治療効果が期待できる肝細胞移植法を確立する。
(2)移植された幹細胞の肝臓内での動態あるいは機能を検討し、ウィルソン病のみならず他の代謝性肝疾患に対する治療として、幹細胞移植が効果的であるか否かを明らかにする。
2.成果
細胞移植実験:移植した細胞の同定を容易にするため、移植の対象として無アルブミンラットを用いた。移植は以下の2つの方法で行った。[方法1]肝上皮細胞をブタ腱由来I型コラーゲンゲルと混合して70%部分肝切除を施した無アルブミンラット肝臓に直接注射した。[方法2]肝上皮細胞をスポンジ状に加工したI型コラーゲン内に播種し、スポシジごと70%部分肝切除を施した無アルブミンラット腹腔内大網上に移植した。
移植後に経時的に採血して、血清アルブミン量の変化をウェスタンブロット法で調べたところ、血清アルブミンの増加を示した無アルブミンラットは、コラーゲンゲルを移植担体に用いた群では67%に、コラーゲンスポンジを用いた群では57%に認められた。移植が成功した例での血清アルブミンの変化を見ると、移植後10週から血清アルブミン量が増加し始め、25週頃まで維持されていた。
本研究において、肝幹細胞としての性質を持った肝上皮細胞をコラーゲンゲルあるいはコラーゲンスポンジを用いて組織工学的に加工して移植した結果、いずれの場合でも無アルブミンラット血清中のアルブミン量が増加したことから、肝上皮細胞が生体内で分化し、肝細胞として機能できることがわかった。このことは、幹細胞移植が代謝性肝疾患に対する治療法として応用できる可能性を示唆するものと考えられる。
