摂食亢進物質オレキシンのカテコールアミン生合成・遊離に及ぼす影響

研究概要	視床下部外及び後側野に主として分布するオレキシンは、1998年に摂食亢進ペプチドとして発見された後、その他多くの生理機能や覚醒行動にも影響していることが明らかになってきた。今回の実験では、オレキシンの中枢神経系以外の作用として、末梢の自律神経特に交感神経系や副腎髄質でのカテコールアミン生合成や遊離に及ぼす影響について検討した。昨年度までの研究結果から、(1)オレキシン-Aは副腎髄質細胞でのカテコールアミン生合成を促進し、その作用点として律速酵素のチロシン水酸化酵素の活性化によるものであること、一方、(2)カテコールアミン遊離には影響しないことが判明した。本年度の研究では、オレキシン-Aの副腎髄質細胞での受容体を同定し、その細胞内シグナル伝達系について検討した。まず、(1)RT-PCR法により副腎髄質細胞におけるオレキシン受容体mRNAの発現を検討した結果、オレキシン受容体1は存在したがオレキシン受容体2のmRNAは確認できなかった。(2)オレキシン-Aのカテコールアミン生合成やチロシン水酸化酵素活性の増加には、少なくともプロティンキナーゼCが関与している可能性が示唆された。以上の結果より、オレキシンは副腎髄質細胞においてオレキシン受容体1-プロティンキナーゼC系を介して、チロシン水酸化酵素を活性化し、カテコールアミン生合成を促進していることが明らかとなった。このオレキシンのカテコールアミン生合成系への影響からオレキシンの脂肪分解やエネルギー代謝への作用について小児肥満や神経性食欲不振症などの栄養障害におけるオレキシンの生理、病理的意義について考察した。
