STAT6 Decoyによる接触アレルギー及びアトピー性皮膚炎の遺伝子療法の試み

研究概要	我々はSTAT6欠損マウスを用いて、接触アレルギーの誘導においてTh2タイプサイトカインであるIL-4,IL-13が重要な役割を果すことを報告した(Yokozeki H et al. : STA6 is essentiall in the Induction of contact hypersensivy. J Exp Med, 191(6) ; 995-1004,2000)。また、最近、接触アレルギーのみならずアトピー性皮膚炎のモデルマウスである抗DNP-IgE抗体投与により誘導される遅発型反応においてもSTAT6欠損マウスでは惹起反応が減弱することを明らかにした。つまり、転写調節因子であるSTAT6を抑制することにより接触アレルギー、遅発型アレルギー反応が抑制しうる可能性があることを明らかにした。今年度はSTAT6Decoyを作成して投与法を確立した。 STAT6DecoyDNA(おとり型DNA)の投与方法の確立 FITCでラベルしたSTAT6 Decoy DNAを作成しHVJ-liposome(共同研究者の大阪大学遺伝子治療学講座金田安史教授より供与)に組み込みBALB/cマウスに皮下、静脈内、腹腔内投与後リンパ節、脾細胞を調整。STAT6 Decoyがリンパ節細胞、脾細胞に取り込めれているかを確認する。また、種々の刺激後リンパ筋細胞、脾細胞の核内蛋白を抽出してGel shift asseyを用いてSTAT6が核内に結合するのを抑制していることを明らかにした。その結果、どの投与法でもリンパ節にDecoyが取り込まれSTAT6の核への結合を阻害していることが明らかにされた。今後、STAT6Deciyの皮下投与法で接触アレルギー、IgE誘導性遅発型反応の制御が可能であるか来年度のに検討予定。
