バキュロウイルス発現蛋白のELISA法を用いた抗デスモコリン自己抗体の研究

研究概要	ヒトDsc1,Dsc2,Dsc3の全長cDNAをtemplateとして用いて、それぞれの蛋白の細胞外部位をコードするcDNAをPCRで単離し、昆虫細胞発現系であるバキュロウイルスのトフンスファーベクターにサブクローニングし、そのC末端にさらにE-tag、His-tagを挿入した。作製した各種のコンストラクトを直鎖状にした後、昆虫細胞であるSF9細胞にトランスフェクトし、相同組み換えにより、DscのcDNAをもったリコンビナントバキュロウイルスを作製し、さらに同様のサイクルを4ー5回繰り返してウイルスを増幅した。最終的に、High-Five細胞を用いてリコンビナントバキュロウイルス蛋白を多量に産生させた。このHis-tagに結合するTalon columnにより培養上清からリコンビナント蛋白を精製した。抗E-tag抗体を用いた免疫ブロット法でリコンビナントバキュロウイルス蛋白が正しく産生されていることを確認した。このヒトDscバキュロウイルス蛋白を用いたELISA法を開発した。すなわち、これらの精製リコンビナント蛋白をELISAプレートにコーティングし、さらに、一次抗体として希釈患者血清と反応後、二次抗体としてperoxidase標識抗ヒトIgGないしIgA抗体と反応させ、発色後、ELISA readerを用いて吸光度を測定した。ELISA法にて陽性所見が得られた血清について、IgA抗体に関しては、Dsc1-3のcDNAトランスフェクション法を用いて、その存在をさらに確認した。ELISA法にて陽性所見が得られた血清について、IgG抗体に関しては、Dsc1-3のバキュロウイルス蛋白を結合させたTalon resinを用いて、Dsc1-3に特異的なIgGをaffinity精製する。その精製IgGを蛍光抗体法で検索し、その特異的染色パターンの有無を検索した。
