メサンギウム細胞特異遺伝子(メグシン)改変マウスにおける腎炎発症機序の解明

研究概要	将来の腎炎に対するゲノム創薬に有用な評価系モデルの確立をめざし、我々が作製した糸球体メサンギウム細胞高発現新規機能遺伝子"メグシン"の高発現マウス(メグシンTg)のin vivo評価系モデルのとしての改良を試みた。ヒトメグシンTgマウスは40週令でヒトのメサンギウム増殖性糸球体腎炎と類似の病理像を自然発症することから、腎疾患予防・治療薬リード化合物のin vivo評価系腎炎モデル動物として有用であることが示されている。しかし、均一な評価系モデル動物としては、より早期に高発症するように改良する必須性がある。その目的でメグシンTgマウスの遺伝子背景改変(C3H, Balb/c)、多重遺伝子改変(MRL/lpr, db/db)、腎障害負荷(片腎摘出)などを試み、腎障害進展の加速を試みた。その結果、1)メグシンTgに片腎摘出を施行すると20週令における糸球体障害率が野生型で2.8±3.1%に対し、メグシンTgで7.3±3.3%と亢進する事が認められた、2)MRL/lpr(自己免疫疾患モデルマウス)との交配で得られたヒトメグシン高発現MRL/lprマウスは、対照のMRL/lprに比してメサンギウム基質への免疫複合体沈着が早期から認められ、それに伴って糸球体障害が有意に加速することが示された、3)メグシンTgマウスの遺伝子背景をC57BL/6、C3H、あるいはBalb/cに変え、各群における腎炎発症率、糸球体障害程度を比較検討してみたが、遺伝子背景による有意な差は認められなかった。また、メグシン遺伝子欠損マウス(メグシンKO)の作製を完了し、遺伝子背景をcongenic化する目的でC57BL/6あるいはBalb/cともどし交配を4世代終了した。
