インスリン開口放出過程における膜蛋白のacylationの意義に関する研究

研究概要

ブドウ糖がKATPチャネルを閉鎖して細胞内Ca^<2+>濃度([Ca^<2+>]i)を上昇させてインスリン分泌を惹起することは良く知られている。一方我々は、従来よりブドウ糖がKATPチャネルの閉鎖とは別のメカニズムでインスリン分泌を刺激しうることを見い出し、その特性や分子基盤について研究してきた。今年度は、膵β細胞からのインスリン分泌、膵β細胞の[Ca^<2+>]iの変動、膵β細胞内のcAMPの定量などを行うことで、次の二点を明らかにした。まず、生理的に重要なインスリン分泌シグナルの一つであるcAMPの増加したラット膵β細胞では、[Ca^<2+>]iの上昇が起こらなくてもブドウ糖がインスリン分泌を惹起しうることを見い出した(Diabetes 51 : S29-32,2002)。さらに、ブドウ糖がインスリン分泌を直接に惹起するのみではなく、何らかの蛋白のアシル化を介して、いわゆる"releasable pool"を増加させることでインスリン分泌に促進的に働いている事実を報告した(J Endocrinol 172: 345-354,2002)。
このアシル化される蛋白の同定が現在の重要な課題であるが、そのため、^3H-パルミチン酸を用いたラット膵β細胞の代謝ラベル実験を遂行中である。現時点では、複数の膵β細胞の蛋白がラベルされることを確認している。現在、様々な条件でこれらのラベルされた蛋白がどのような挙動を示すのか詳細な検討を加えるとともに、二次元電気泳動でこれら蛋白の同定を試みている。
