LDL受容体欠損家兎におけるin vivo遺伝子修正:標的相同組換えと受容体を介した遺伝子導入による新たな遺伝子治療の試み

研究概要	ウイルスベクターを用いる遺伝子導入は安全性に問題がある。我々はウイルスベクターを用いずに、新たに開発されたリポソームにリガンドを結合させ、受容体を介して遣伝子を導入する安全な技術を改良した。一方、家族性高コレステロール血症は著明な高コレステロール血症を呈し、若年から冠動脈疾患を惹起する重要な疾患であり、LDL受容体欠損が原因であることがすでに明らかにされている。我々は、本疾患のモデル動物であるWHHL家兎に新たに開発した方法を用いてLDL遺伝子の導入を試みた結果、導入に成功した。合成陽性脂質である(+)-N, N[bis(2-hydroxyethyl)-N-methyl-N-[2,3-di(tetradecanoyloxy)propyl]ammonium iodideと中性脂質であるDOPEからなるリポソームに対して、trans ferrinのリガンドを結合させた後、rabbit LDL受容体cDNAを高発現プロモータを含むベクター(pTARGET)に結合させたconstructを用いてリポソームリガンドDNA複合体を作成した。これをWHHL家兎に静注投与した後、血漿LDLコレステロール値を経時的に測定した。LDL受容体cDNA投与量は50μgとした。 LDL受容体cDNA50μg静注投与により、最大10%の低下と3週間の効果持続を確認した(Gene Therapyに掲載予定)。また、リンパ球におけるLDL受容体遺伝子発現を確認した。今後、研究をさらに発展させて、LDL受容体cDNA投与量の増加および繰り返しの投与により、LDL受容体遺伝子発現を増加させ、LDLコレステロール低下作用の増加および維持が可能であるか確認したい。
