研究概要	アクアポリン・アディポース(AQPap)は脂肪組織特異的に発現するグリセロールチャネルである。マウス及びヒトAQPap遺伝子のプロモーター領域に核内受容体型転写因子PPARγ(peroxisome proliferator activated receptor)が結合するPPRE(peroxisome proliferator response element)を認めた。この転写調節機構によりAQPap遺伝子は脂肪組織特異的に発現することを示した。AQPapがインスリンにより抑制されることを示した。7塩基配insuli negative response elementをAQPapのプロモーター領域に認めた。 AQPapのヒト疾患に対する意義を解明するため、ヒトAQPap遺伝子の構造を明らかにした。160人の遺伝子解析を行い、3つのミスセンス変異(R12C, V59L, G264V)と2つのサイレント変異(A103A, G250G)を見出した。それらAQPapヘテロ変異と肥満及び糖尿病との関連性は見られなかった。同定したAQPap遺伝子変異蛋白の機能解析をXenopus oocytesを用いて行った。AQPapG264V蛋白はグリセロール透過能を欠失していた。AQPap-G264Vホモ変異を持つ患者は、AQPap-wildと同様に運動負荷によりノルアドレナリン濃度が上昇するにもかかわらず、血中グリセロール濃度上昇に関連することが示唆された。現在、F1マウスが誕生してきており、今後交配により個体数を増やし、ヘテロ型とヘテロ型との交配で誕生したマウス(野生型、ヘテロ型、ホモ型)の経時的な体重変動や代謝パラメーターを観察し、同時にマウスの遺伝的背景をより均一化するためにbackcrossを進め、生体内におけるグリセロール代謝を中心としたAQPapの生理的・病態的意義を明らかにしていく予定である。
