CD4陽性T細胞に認識される上皮性癌抗原遺伝子の同定と癌ワクチンへの応用

研究概要

クラスI分子に提示される癌抗原ペプチドを用いた抗癌ワクチン療法の治療効果を増強することを目的として、CD4陽性ヘルパーT細胞に認識されるヒト癌細胞の癌抗原をコードする遺伝子を新たに単離することを、また、既知の癌抗原に関しては、クラスII分子に提示されCD4陽性T細胞に認識される新たな抗原ペプチドを同定することを試みた。卵巣癌患者の癌性腹水よりクラスII分子を恒常的に発現しているアロ腫瘍であるB細胞系・単球系白血病細胞を自己HLA-DRB1^*04051拘束性に認識するCD4陽性T細胞(SA-1)を確立した。このCD4陽性細胞は自己末梢リンパ球にも反応し自己反応性を有することが判明した。cDNAライブラリー発現クローニングを行なうために、HLA-DRA1^*0101,-DRB1^*04051を恒常的に発現した293T細胞(293T-DR0405)を確立し、また、細胞内にクラスII抗原プロセッシングに必要な分子(IiやHLA-DM)を発現させるためにCIITA cDNAを準備しtransfectしたが、SA-1細胞はCIITAをtransfectしない293T-DR0405細胞にも反応した。また、クラスII分子を発現した上皮性癌細胞株を反応性確認のスクリーニングに用いることを目的に、CIITAを恒常的に発現した(内因性クラスII分子を発現することを意味する)上皮性癌の細胞株を作製した。さらに、既知の癌抗原で抗体が認識することを確認しているcyclophilin BとART4抗原のHLA-DRB1^*1501分子に結合が期待されるクラスIIペプチドを用いて健康人の末梢リンパ球をdendritic cellsを用いて刺激し、クラスIIエピトープの同定を試みたが、自己反応性CD4陽性T細胞の優位な増加のため未だ決定に至っていない。
