CD4陽性T細胞に認識される上皮性癌抗原遺伝子の同定と癌ワクチンへの応用

研究概要

1.申請者は上記科学研究費補助金により、卵巣癌患者の癌性腹水よりクラスIIを恒常的に発現しているアロ腫瘍であるB細胞系・単球系白血病細胞を自己HLA-DRB1^*04051拘束性に認識するCD4陽性T細胞(SA-1)を確立した。それらはHLA-DRB1^*04051を発現している大部分の細胞、特に293T-DRB1^*0405細胞にも反応することからcDNA library expression法によるクローニングが困難であった。しかしながら、細胞内のクラスII pathwayに必要な分子であるinvariant chain (Ii) やクラスIIトランスアクチベーター(CIITA) plasmid cDNAを準備することができた。
2.久留米大学では、HLAクラスI分子に提示される癌抗原ペプチドを癌患者に投与する癌ワクチンの臨床治験が準行中であるが、患者によっては投与された癌抗原ペプチド(9-mer,10-mer)に対するIgG抗体が著明に誘導される場合があることを観察している。生体内で抗原特異的IgGが誘導されるためには、抗体を産生するB細胞にサイトカインを供給するヘルパーT細胞の存在が示唆されるが、実際に9-merであるUBE2V 43-51ペプチドを投与された肺癌患者の末梢リンパ球中に、UBE2V 43-51ペプチドをHLA-DRB1^*0403拘束性に認識するCD4陽性T細胞の存在を確認した。また、UBE2V 43-51ペプチドは6番目にシステインを有しており、酸化によりシスチン化や2量体化されやすいが、UBE2V 43-51ペプチドを認識するCD4陽性T細胞は、BE2V 43-51ペプチドを単量体としてのみでなく2量体としても認識することを証明した。
3.申請者は、再燃前立腺癌に対するワクチン療法確立のため、日本人に多いHLA-A^*2402分子に提示され前立腺癌に対する細胞傷害性T細胞を誘導できるペプチド同定を試み、prostate-specific antigen(PSA)152-160,PSA24-257ペプチドを同定した。また、PSA 248-257ペプチドは、前立腺癌患者の約60%、健康人の約80%でこのペプチドに対するIgG抗体の存在が認められた。このことは、前立腺癌患者や健康人にPSA抗原を認識するCD4陽性T細胞が存在することを間接的に示唆していると考えられ、現在この可能性を検討中である。
