肝阻血再灌流障害の遺伝子治療による制御と肝再生促進に関する研究(特に転写因子レベルでの検討)

研究概要

1.遺伝子導入による制御に関する検討
1)遺伝子導入に関しては未だ導入効率が悪い状況であり今後さらに検討が必要であると考えられた。
2)STAT6とNFκBのcoactivatorであるCBPに関しては免疫沈降による検討では現時点では、明かな結果が得られずさらに検討が必要と考えられた。
肝阻血再灌流障害の制御に関して、real-time PCRによりCC chemokineの発現の検討を行った。再灌流後経時的にCC chemokineの発現を認め病理組織標本での検討では好中球以外に単核球の集積があることが認められた。またELR motif negativeのCXC chemokineも再灌流後発現しており、様々なmediatorが発現し障害を修飾していることが判明した。(肝臓 2002;43:A194)
また内因性に障害を制御する機構としてIL-48に注目しその発現と抗体投与による障害への関与を検討した。その結果IL-18は抗炎症性サイトカインの発現を抑制し障害を増悪することが判明した。(日外会雑誌 2002;103:198)
2.Ischemic preconditioningに関しては、至適時間が決定できこれにより明らかに長時間の阻血再灌流障害が軽減されるという結果が得られた(P<0.01)。さらに組織学的好中球集積やchemokineの発現がreal time PCRによる検討でmRNAレベルで有意に抑制されるという結果が得られた。今後さらにその機序に関して転写因子レベルで検討を進めるところである。
さらに阻血中に発現するmediatorに関してはmacrophage migratory inhibitory factorの発現があることが免疫組織科学染色並びにmRNAレベルで判明した。
肝再生促進実験に関しては、今後検討を進める予定である。
