食道癌細胞におけるProtein Kinase CとChimaerinの役割

研究概要	(目的)食道癌細胞におけるprotein kinase C(PKC)とβ2Chimaerinの役割を分子生物学的手法を用いて検討した。(結果)食道癌培養細胞株KE3にPKCのactivatorであるPMAを30nM、1時間投与すると投与24時間後にはcontrolと比較しで23%の増殖抑制作用が認められた。細胞周期の検討ではPMA投与群ではcontrolと比較してS、G2/M期が14.6%増加した。この結果よりKE3の細胞増殖にはPKCが関与していることが考えられた。さらにKE3の内因性PKCδはwestern blotの解析では中等度の発現が認められた。従って、KE3細胞においてPKCの中でもPKCδはKE3細胞の増殖に関与している可能性が考えられた。この現象を裏付ける目的でKE3にPKCδ adenovirus(PKCδ AdV)を感染させ、PKCδを強制発現させることによる生物学的効果を検討した。PKCδ AdV感染24時間後にPKCδ蛋白の発現をwestern blotで確認したが、良好な発現が認められた。感染後24時間後にはcontrolと比鞍して60%の細砲増殖抑制が認められた。さらに、感染後にPMAでPKCδをactivateすると73%の細胞増殖抑制が認められた。以上より、KE3細胞においてPKCδは強い細胞増殖抑制作用を有することが示唆された。食道癌培養細胞株8種類を使用し、β2Chimaerinの発現を検討するために、RT-PCRを施行した。KE3、KE4、KE5、KE6、KE7、KE8、KE9の7種類の細胞株ではβ2chimaerinの軽度の発現を認めており、切除標本を用いた免疫染色の結果からも上皮細胞の増殖に何らかの関係があるものと考えられた。(今後の予定)β2chimaerinの生物学的機能を検索する目的でβ2chimaerin遺伝子を組替えたadenovirusを作製した。β2chimaerin adenovirus感染後に良好な蛋白発現が認められた。今後、chimaerin強制発現後の生物学的効果を明らかにしていく。
