虚血脳保護作用を有するサイトカインの細胞内情報伝達機構

研究概要	我々はこれまでに、インターロイキン3(IL-3)やエリスロポエチン(EPO)、またはEpidermal growth factor(EGF)が脳虚血に対して保護的に働く事を報告してきた。同時にIL-3やEPOでは細胞死抑制遺伝子産物Bcl-X_Lの発現増強を介して神経細胞を強力に保護すことも報告した。IL-3、EPO、EGFはいずれもJak/Stat系を介して細胞内シグナル伝達を行うことが知られており、Stat3やStat5によりBcl-X_Lが発現誘導され抗アポトーシスに働く事も知られている。そこで我々はまず、脳虚血時に実際にJak/Stat系の活性化が起こるかどうかをStatのリン酸化抗体を用いて検討した。その結果Stat3のリン酸化が脳虚血後6時間目から虚血巣で著名に亢進する事を認めた。更に免疫電顕によりリン酸化Stat3陽性細胞はすべて変性神経細胞である事を示し、Stat3の活性化は細胞死に関連する可能性を報告した。次いでStat3を組み込んだアデノウイルスベクターを作成し、初代培養神経細胞やグリア細胞にstat3のwild type(st3.Wt)やそのdominant negative type、及びレポータ遺伝子(LacZ)を強制発現させ、その効果を検討した。その結果初代培養グリア細胞にSt3.Wtを強制発現させると明らかに細胞死が促進される事が分かった。そして細胞死に陥る直前にはグリア細胞でのbcl-xlの発現が低下している事とcaspase-3活性が上昇している事を示した。一方神経細胞に上記のウイルスベクターを用いて強制発現させても有意な変化は認められず、bcl-xlの減少も認めなかった。しかしながら強制発現後の神経細胞をマイクログリアと共培養すると、共培養後3日目にはSt3.Wtを強制発現させたニューロンで細胞死の促進効果が見られた。
