胚発育に対する至適子宮内膜環境の設定:子宮内膜線上皮細胞と間質細胞の相互作用の観点から

研究概要

着床のための子宮内膜の機能的成熟には、腺上皮細胞と間質細胞の相互作用が重要であることを明らかにするため、両者の細胞を共培養することによってそれそれの細胞の単独での培養と比べ、細胞の機能が変化するかを検討している。前年度の研究により、着床に不可欠な因子であるLeukemia Inhibitory Factor(LIF)の上皮細胞における発現は、estradiol(E;10^<-8>めが間質細胞に作用し、何らかの物質の産生を介し、増加することが明らかとなっている。1.今年度は、Eによって間質細胞から分泌されて上皮細胞のLIF発現を増加させる因子としてinsulin-like growth factor-1(IGF-1)に注目した。子宮内膜から間質細胞と上皮細胞を分離し、間質細胞は6ウエル培養プレートに定着させ、上皮細胞は、マトリゲルをコートしたインサート上に定着させた。間質細胞のウエル内に上皮細胞が定着したインサートを入れ相互作用ができるようにした後、培養液中に抗IGF-1抗体を添加し、Eの存在下に3日間培養したところ、Eによる上皮細胞のLIF発現増加作用は、完全に阻止された。すなわち、Eが間質細胞に作用し、IGF-1産生を介し、上皮細胞からのLIF産生を増加させていることが明らかとなった。2.今度は逆に、間質細胞の機能的成熟における腺上皮細胞の作用を検討するため、上皮細胞が間質細胞の脱落膜化に及ぼす影響を検討した。上記のように、両者の細胞を相互作用ができるようにし、Eとmedoroxyprogesterone(MPA;10^<-6>M)の存在下で12日間培養後、間質細胞の形態変化と脱落膜化の指標であるinsulin-like growth factor-binding protein-1(IGFBP-1)の発現を調べた。EとMPAにより間質細胞は形態的に脱落膜変化を示した他、IGFBP.1発現も有意に増加したが、上皮細胞の存在下では、この脱落膜化が有意に抑制された。したがって、上皮細胞は、間質細胞の脱落膜化を抑制することによって、implantation windowの閉鎖を遅らせ胚受容が可能な期間を延長させていることが示唆された。
