マクロファージ機能の制御による実験的ぶどう膜炎抑制モデルの作成

研究概要

平成14年度も、ぶどう膜炎の抑制にMCP-1を標的とすることが可能かを研究する目的で、1)MCP-1 Tgmを用いた実験的自己免疫性ぶどう膜炎(EAU)マウスモデル、2)C57BL/6(B6)マウスにおける抗MCP-1抗体によるぶどう膜炎の発症抑制の試み、について解析した。
EAUはTgm、non-Tgm(B6の背景)あるいはB6の頚部背側、腰部背側にhuman interphotoreceptor retinoidbinding protein peptide 1-20+完全フロイントアジュバント100μ1を投与し、同日百日咳毒素0.1μg/マウスを腹腔内投与することによって誘導した。翌日よりThurauらの基準に従ってスコア化(0〜4)、眼球は剖検後に病理組織学的評価(0〜4)を行った。抗MCP-1抗体投与は5μg/マウス/日で、連日(3週間)行った。その結果;
1)EAUのピーク時に、Tgmでnon-Tgmより高いEAUスコアを示した。
この現象は、MCP-1がぶどう膜炎発症抑制の標的となり得ることを示唆した。そこでB6マウスにEAUを誘導し、抗マウスMCP-1抗体投与によって発症抑制が可能であるか検討したところ;
2)抗体投与群でIRBPp1-20に対し強いT細胞応答および高いサイトカイン(TNF-α,I FN-γ,I L-5)産生を呈した。亢進したT細胞応答に呼応して高い病理組織スコアを示した。
MCP-1を抗体で感作当初よりブロックすることはぶどう膜炎治療の目的にはそぐわないことが判明した。実験1)と2)の結果は矛盾するが、1)についてはMCP-1の構成的高発現により下流のシグナルが伝達されないことがTgmでのEAU悪化と考えると、両者の結果は一元的に理解できる。上記の現象にはTNF-αの産生亢進が基礎にあり、抗TNF-α抗体療法の基盤を考える上で重要な知見と考えられた。
