一次視覚路におけるアセチルコリン神経の証明と機能解析

研究概要	視覚情報を網膜から脳に伝える網膜神経節細胞の神経伝達物質に関しては、依然として確立していない。カエルの網膜においてはアセチルコリンがその候補として挙げられたことがあるが、一般的には網膜神経節細胞の神経伝達物質としてのアセチルコリンの役割については否定的である。一つには、従来アセチルコリンの合成酵素であるコリンアセチル基転移酵素(ChAT)の抗体では網膜神経節細胞が標識されないことがその理由であった。近年、我々はChATの新しいスプライス異型を同定し、pChATと命名した。本研究課題では、pChATの発現を指標にして網膜神経節細胞の神経伝達物質がアセチルコリンである可能性を追求した。本研究により下記の点が明らかとなった。 1.免疫組織化学、及び逆行性トレーサーとの2重標識により、ラットの網膜神経節細胞の約20%がpChAT陽性であることを明らかとなった。 2.ウェスタン・ブロット法とRT-PCR法により、網膜におけるpChATタンパク及びそのmRNAの発現が確認された。 3.網膜と視神経を用いたChAT活性の測定により網膜神経節細胞の中にコリン作動性の細胞が含まれることを明らかにした。 4.一次視覚路のおけるpChAT発現は明暗の状況により影響された。以上の結果は、一次視覚路におけるアセチルコリンの役割を示唆するものとして重要であり、J Neuroscience誌に発表した。さらに、pChATを指標にすることにより、脳の睡眠・覚醒に関与する神経細胞群に新たなコリン細胞を見い出すと共に(Neuroscience)、虹彩のコリン神経支配についても新たな視点を加えることに成功した(論文投稿中)。
