アクアポリン5変異蛋白質発現用培養唾液腺細胞の作成と水分泌機構解明へのアプローチ

研究概要

本研究では水輸送に関与する蛋白質アクアポリン(AQP)の内、外分泌腺型AQPであるAQP5によるトラフィッキングおよびAQP5機能の調節機作を明らかにし、開口分泌に関与しているSNARE蛋白質とAQP5の関係、および加齢に伴うAQPとイオンチャネルならびに各種輸送体との共役メカニズムを明らかにすると共に、これらの複合調節を究明することを目的とした。本年度は、ラットAQP5による水輸送の分子メカニズムとその機能調節の研究を行うため、まず、細胞質ドメイン(ループD)に存在するPKAターゲットモチーフの役割を究明する事とした。すでに教室においてクローニングしたラットAQP5 cDNAの当該配列のうちセリンをアラニンに変異させたもの、スレオニンをバリンに変異させたもの、およびセリンとスレオニンの両方をそれぞれアラニンとバリンに変異させたものをin vitro mutagenesisよって作成した。変異を導入しない野生型と合わせて4種のcDNAを個別にヒグロマイシン耐性遺伝子を含むベクターに組み入れ、極性を示すMDCK細胞に遺伝子導入した。MDCK細胞はあらかじめクローニングし、均一化した株をもちいた。ヒグロマイシン選択培地で生育した4種の遺伝子導入細胞はそれぞれ野生型、変異型AQP5を発現していることをウェッスタンブロッティングにより確認した。細胞の免疫染色を行うと野生株およびセリン、スレオニンのいずれかを変異したAQP5遺伝子を導入された細胞では細胞内と細胞膜の両方にAQP5蛋白質は分布したがセリン・スレオニンダブルミューテーションによりAQP5蛋白質の細胞膜への局在は抑制されている事を予備実験により見いだした。14年度はこの事実をふまえ、トラフィッキングにおけるAQP5/PKAターゲットモチーフの役割をさらに追求する。
