アクアポリン5変異蛋白質発現用培養唾液腺細胞の作成と水分泌機構解明へのアプローチ

研究概要	本研究では水輸送に関与する蛋白質アクアポリン(AQP)の内、外分泌腺型AQPであるAQP5によるトラフィッキングおよびAQP5機能の調節機作を明らかにすることを目的とした。 1.ラットAQP5による水輸送の分子メカニズムとその機能調節の研究を行うため、まづ、細胞質ドメイン(ループD)に存在するPKAターゲットモチーフの役割を究明する事とした。前年度までに、教室においてクローニングしたラットAQP5 cDNAの当該配列のうちセリンをアラニンに変異させたもの、スレオニンをバリンに変異させたもの、およびセリンとスレオニンの両方をそれぞれアラニンとバリンに変異させたものをin vitro mutagenesisよって作成した。本年度はさらにN端またはC端にGFPを導入したAQP5 cDNAを作成した(GFP-AQP5およびAQP5-GFP)。これらのrAQP5はMDCK細胞およびHSG細胞に遺伝子導入し、各種シグナル伝達薬のGFP-rAQP5局在に対する影響を調べる。 2.STR/Nマウスは高齢化により多渇多飲となる。本マウスにおける腎AQP2 mRNAは多飲の発症とほぼ同時に減少た。生後10週齢では飲水量は薬30%増加したが腎AQP2 mRNAはすでに正常マウスの20%に減少した。生後45週齢では飲水量は正常マウスの5.5倍に達し、腎AQP2 mRNAは正常マウスの10%になった。バソプレッシンレベルはSTR/Nマウスではむしろ微増するとされているので腎AQP2 mRNAの減少は多尿に基づくフィードバック調節が存在する可能性が示唆された。 3.胎生期唾液腺におけるAQPs(AQP1-5)の発現を検索した。RT-PCRの結果、胎生期顎下腺ではAQP1,3,4,5の発現が認められた。ノーザンブロットによりAQP5は胎生16日から19日にかけて徐々に発現量が増加し、いごは強く発現したが、AQP1は胎生期を通じほぼ同レベルで発現した。これらの結果はRT-PCRの結果とも一致した。一方、AQP3、4はノーザンブロットでは検出できず、発現量は極めて低いと考えられた。生後においても成長に関わらず、AQP5とAQP1はほぼ同レベルで発現するが、AQP3、4は検出されなかった。いずれの発生段階の顎下腺においてもAQP1は血管に、またAQP5は腺房細胞の管腔膜に局在したが、後者は腺房部分泌細管や神経繊維及一部の血管にも局在した。以上の結果およびAQP1ノックアウトマウスの結果より、唾液の産生と分泌は発生過程を通じてAQP5によって調節されると考えられた。
