細胞交通を考慮した接触皮膚炎の感作過程の分子論的解析とその制御法の開発

研究概要	接触皮膚炎の動物モデルにおいて、その感作時に皮膚真皮からリンパ節へ移動するマクロファージ様細胞に着目した研究を行っている。感作という事象を(1)マクロファージ様細胞の遊走開始、(2)リンパ節の特定の領域への分布と定着、(3)低分子抗原(ハプテン)の提示過程におけるマクロファージの相互作用、の3つの段階に分割し、それぞれの素過程を連続したものとして捉えることから、感作過程の理解を目指している。真皮マクロファージ様細胞のマーカーであるマクロファージレクチン(MGL)が機能的な認識分子として体内で細胞交通を規定する可能性を示した。皮膚からの細胞が最初に流入する部位はリンパ節の辺縁洞である。この部分に存在する辺縁洞マクロファージが組換え型MGLと高い親和性で結合することを見出した。リンパ節の可溶化物からMGLに結合性を示すリガンド分子の同定を行ったところ、これがsialoadhesinであることが強く示唆された。さらに生化学的解析からMGLとsialoadhesinの結合にN結合型糖鎖が関与すること、細胞膜上に発現したMGLがsialoadhesinと結合することを示した。真皮マクロファージと辺縁洞マクロファージの相互作用の分子基盤の一つを明らかにした。ハプテンを塗布したリンパ節からリンパ節細胞を取得し、MGLを発現する細胞の性質を表面抗原に着目して解析した。この細胞は高頻度にハプテンを保持していること、樹状細胞のマーカーであるCD11cや共刺激分子CD86を発現していることを明らかにした。この事実はMGL陽性細胞がリンパ節において抗原を実際に提示する細胞としてふるまうことを示唆している。マウスの系統間で比較すると、ハプテンを保持したMGL陽性細胞の割合は感作の大小と必ずしも相関しなかった。今後この細胞の感作過程における性質をさらに検討する必要がある。
