サブタイプ特異性の高い新規グルタメイトレセプター内在性リガンドの探索研究

研究概要	グルタメイト(Glu)レセプターには、少なくとも22種類の異なるサブユニットが存在するが、これらサブユニット群の異なる組み合わせから構成されるレセプター蛋白質が、脳内特定部位に無数に分布すると予想される。今年度は、特にシグナル伝達および神経毒性発現に関連の高いGluトランスポーターの脳内多様性について、オートラジオグラフィー法を用いて検討した。ラット脳から凍結水平切片を切り出したのち、4℃で20nM[^3H]L-aspまたは[^3H]D-aspと反応させた。反応終了後、得られた切片をトリチウム感受性フィルムに、トチリウム標準線源とともに感光させた。両放射性基質を切片と反応させた結果、どちらの場合も脳全体に結合が見られたが、海馬や大脳皮質などの前脳部位に比べ、小脳灰白質で2倍から3倍の強い結合が観察された。さらに、両放射性基質の結合を比較すると、前脳部位では[^3H]L-aspの特異的結合は、[^3H]D-aspよりも1.2倍から2.3倍強力であった。一方、小脳灰白質では、[^3H]L-aspの結合は[^3H]D-aspよりも2.2倍から3.3倍強力であり、より顕著な結合能の違いが観察された。[^3H]L-asp結合に対して、L-aspもD-aspも大脳皮質、海馬、線条体および中核の前脳部位では、ほぼ同等度の阻害活性を示した。これに対して、小脳ではL-aspはIC_<50>値441nMと他の脳内部位と同程度の阻害活性を示したが、D-aspは1386nMとL-aspと比較して3倍以上弱い阻害活性を示すことが判明した。同様に、[^3H]D-aspに対しても前脳部位では両異性体はほぼ同程度の阻害活性を示したが、小脳ではD-aspはL-aspよりも3倍以上弱い阻害活性を示した。以上の結果から、小脳に発現するグルタミン酸トランスポーターはL-aspとD-aspを識別する性質を有する可能性が示唆される。
