プロスタグランジンの標的ニューロンにおける発熱関与遺伝子の同定と機能解析

研究概要	発熱反応は、末梢からサイトカインなどを介して視床下部の体温中枢にシグナルが伝達された結果、引き起こされる中枢性の応答である。申請者らは、PGE受容体の4種類のサブタイプを個々に欠損したマウスの作成し、EP3受容体欠損マウスが炎症性の発熱を全く示さないことを明らかにした。本研究の目的は、(1)EP3を発現する視索前野ニューロンの個々に関して、それぞれどの様な神経伝達物質、チャネル、受容体を発現するのか解析し、EP3発現ニューロンの基本的な性質を同定すること、(2)EP3ニューロンの個々において、末梢からの発熱シグナルの有無によりどの様な遺伝子の発現が変化するのかを調べることで、発熱刺激時にEP3受容体のすぐ下流で働く遺伝子群を検索することである。本年度、申請者は、ペンシルバニア大のEberwineらによって開発されたシングルセル遺伝子発現プロフィール解析法を国内において初めて確立した。また本解析法を用いることにより、視床下部のEP3ニューロンに共通して発現の見られる遺伝子群を同定した。現在、これらの候補遺伝子群が視床下部において本当にEP3と共存しているのか検討中である。また、動物実験における個体差のように、細胞間のゆらぎが問題となったが、複数の個体から複数の細胞を解析に供することで、生物学的な差を捉えることが可能であった。さらに、本法を種々の試料に対しても対応できるように最適化し、またハイスループット化を検討する予定である。
