ヨードシンポーターによる実験的自己免疫性甲状腺炎・唾液腺炎の誘導に関する研究

研究概要	橋本病患者の約2割は角結膜炎、口内乾燥症などのシェーグレン症候群を合併するが、なぜ合併するのか全くわかっていない。ヨードシンポーター(NIS)が甲状腺のほか唾液腺にも発現していることに注目し、NISをマウスに免疫することにより、甲状腺炎および唾液腺炎を発症するかを調べ、ヒトにおける自己免疫性甲状腺炎(橋本病)および自己免疫性唾液腺炎(シェーグレン症候群)、.特に両者の合併例におけるNISの病因的役割を明らかにすることが本研究の目的である。平成12年から13年度の科学研究費補助金により作成した、NISを高発現した293細胞をmitomycin C処理で増殖しないようにした後、Complete Freund's Adjuvantと混和し、CBA/Jマウスそけい部の皮下に投与したグループをNISを発現していない293細胞で免疫したコントロールグループと比較し検討した。予想に反し、NIS発現細胞で免疫したマウスでも甲状腺および唾液腺にリンパ球浸潤などの炎症所見が認められなかった。NISに対する自己抗体をradioligand assayおよびNIS発現3T3細胞でのヨードの取り込み阻害活性で検討したが検出できなかった。甲状腺ホルモン値も有意な変化を認めなかった。以上のことよりNIS発現細胞を用いた免疫方法では自己免疫反応を起こすには十分ではないと判断した。近年、DNAで直接マウスをを免疫するGenetic Immunizationが行われつつある。NIS同様膜たんぱくであるため、自己免疫反応を誘導しにくかったTSHレセプターをGenetic Immunizationすることにより甲状腺炎を誘導できたとの報告が最近された。今後、NISをGenetic Immunizationすることにより、甲状腺炎および唾液腺炎が誘導できないか検討する予定である。
