研究概要	(1)L型Ca^<2+>チャネル阻害剤ω-コノトキシンTxVIIとNa^+チャネル不活性化阻害剤δ-コノトキシンTxVIAは3本のS-S架橋による4ループ構造をもっており、まずそれらの前半と後半を入れ替えた2種のキメラアナローグδδωωとωωδδの合成を試みた。ωTxVIIに有効であった条件で空気酸化によるS-S架橋形成をおこなったところ、δδωωはωTxVIIと同程度の収率で目的物が得られたが、ωωδδの場合はHPLC分析で主要なピークは認められなかった。環化条件をδTxVIAに有効であったものに変えたところ、ゲルろ過後の単量体画分に質量が理論値と一致するものが主成分として含まれることが明らかとなった。これらの結果からωTxVIIの後半部分にS-S架橋形成に重要な残基があると考えられた。現在、単量体画分からの目的物の単離および、より有効な環化反応条件の検討を行っている。 (2)筋肉のNa^+チャネルを選択的に阻害するμ-コノトキシンGIIIAのC末端Alaを酸性側鎖をもつGluに置換したアナローグ(A22E)を合成した。電気生理学的解析によりA22Eは天然GIIIAより約90倍弱い活性しか示さず、Na^+チャネルに結合した際にはGIIIAのC末端が相互作用する近傍に酸性残基があるものと予想された。変異したNa^+チャネルを用いて系統的に調べたところ、GIIIAのC末端はチャネルの第2ドメインに近接することが示唆された。 (3)東南アジア産のサソリから単離された新規のペプチド毒HSトキシン5種の合成をおこなった。その中で分子内に2本のS-S架橋をもつ4種の毒についてはC末端アミド体と遊離カルボキシル体を合成し天然物との比較によりC末端部分の構造を確定した。
