核内レセプターを介する新規細胞内シグナリング機構の解析

研究概要	我々は、核内レセプターFXR(Famesoid-X-Receptor)が胆汁酸に応答して、ヒト胆汁酸トランスポーターI-BABP遺伝子を転写活性化することを明らかにした。更に、1-BABP遺伝子の転写制御機構を詳細に解析したところ、胆汁酸によるI-BABP遺伝子を転写活性化は、ステロール存在下で抑制され、ステロール生合成阻害剤の共存下で促進されたことから、SREBP(Sterol Response Element-Binding Protein)の関与が不唆された。プロモーター解析の結果、SRE様の配列(-72/-62)及びこの配列に隣接して、SREBPのコファクターとして働く転写因子Sp1が結合するccBox様配列(-58/-48)があり、これらの配列は、いずれもステロールによる抑制に関与し、胆汁酸応答配列(BARE=FXRE)と同様に種を超えて保存されていることが確かめられた。これは、胆汁酸で可溶化されたコレステロールの濃度が高いときは、I-BABPの発現を抑制し、胆汁酸の腸肝循環の効率を下げることによって、余分のコレステロルをコレステロールトランスポーターABCA1を介して体外へ排出しているのかもしれない。興味深いことに、I-BABP遺伝子は核内レセプターLXR(Liver X receptor)のアゴニストによって転写活性化されたが、この活性化には、LXRの標的遺伝子SREBP-1cの関与が示唆された。このように、ヒトI-BABPの遺伝子発現は核内レセプーFXR、LXRおよびSREBPなど、クラスの異なるリガンド応答性転写制御因子によって厳密に制御されていた。従って、I-BABPは腸肝循環における胆汁酸の代謝および生体のコレステロールのホメオスタシス維持において重要な機能を果たしていることが確かめられた。また、FXRは、肝臓、小腸、腎臓以外、大腸、副腎、卵巣、精巣、脳などの組織でも発現していることから、FXRの標的遺伝子はコレステロールのホメオスタシス維持だけでなく、高次生命現象の制御にも関与している可能性がある。
