研究概要	RhoファミリーGTP結合蛋白質に対するグアニンヌクレオチド交換因子(GEF)であるDb1ファミリー蛋白質は、細胞骨格系の再構成など、種々の細胞機能を調節している。本研究課題では、上流からの刺激に応答したDb1によるRhoファミリーの活性調節機構を解析した。Db1は、Cdc42の標的分子であるACK-1チロシンキナーゼによりリン酸化され、その結果、RhoA、Rac1、Cdc42に対するGEF活性が上昇することが知られている。一方ACK-1は、アダプター分子Grb2を介して上皮成長因子レセプター(EGF-R)と会合するので、EGF-Rの下流でのDb1の機能を検討した。その結果、EGF刺激によって、ACK-1とDb1のチロシンリン酸化が誘導され、その際、EGF-Rの下流でCdc42、Grb2が関与することが明らかとなった。これに伴って、Db1が活性化され、RhoA、Rac1、Cdc42を介して、アクチン細胞骨格系の再構成が誘導された。以上の結果より、チロシンリン酸化によるDb1の活性調節が、EGF-Rの下流で実際に機能していることが強く示唆された。一方、Db1の近縁分子には、多くのスプライスバリアントが存在することも明らかとなった。現在、活性調節に関与すると考えられるドメインの機能を検討しており、今後それらの特異的な生理機能を明らかにしていく予定である。一方、Rasファミリーに関しては、まずRasあるいはRap1の標的分子として機能するPLCεのRap1 GEFドメインが、ゴルジ装置におけるRap1の下流での持続的活性化に必要であることが示唆された。また、他のRap1 GEFであるRA-GEF-1とRA-GEF-2は、それぞれ、Rap1、M-Rasの下流で、ゴルジ装置あるいは細胞膜に存在するRap1を介するシグナル伝達系の増強に関与していることを示した。
