自然免疫におけるTLR4を介したリポ多糖(LPS)認識と活性化の分子機構

研究概要	細菌内毒素(エンドトキシン)として知られるグラム陰性菌表層のリポ多棟(LPS)をはじめとした様々な細菌菌体成分は、Toll-like receptor(TLR)を介して細胞応答反応を惹起し、自然免疫系を活性化する。一方、これら菌体成分によるTLR活性化の分子反応については、未だほとんど明らかでない。本研究では、TLR4をはじめ、他の様々なTLRの細菌菌体成分による活性化の分子機構を明らかにするため、以下の研究を行った。すでに研究代表者が見出していた、培養細胞に強制発現したTLR4を免疫沈降した際に、TLR4と共沈殿するタンパク質について、質量分析計を用いた内部ペプチド断片の解析を行い、その同定に成功した。現在、このタンパク質のTLR活性化反応における生理的意義について検討しつつある。一方、TLR活性化には、細菌菌体成分によってTLRに結合する内在性リガンドが存在するという仮説にもとづき、リガンドの同定に向けた計画を実行しつつある。現在までに、リガンド同定のプローブとして使用する目的で、TLR2、TLR4の細胞外ドメインをイムノグロブリンのFc領域と融合させた融合タンパク質について、昆虫細胞を用いた発現系を確立した。この発現系を用いて発現した昆虫細胞の培養上清中のTLR2-Fc融合タンパク質を、Protein A-Sepharoseを用いて単一成分まで精製した。この組換タンパク質について、ペプチドグリカンによるTLR2活性化反応に及ぼす影響をTLR2遺伝子導入細胞を活用したNF-κB reporter assayによって検討したところ、培養液に加えた組換タンパク質は、刺激時のTLR2活性化を阻害することが明らかになり、生理活性をもつタンパク質であることが確認された。今後、これらの融合タンパク質に結合する因子の検索を行う予定である。
