セレクチンと糖鎖を介した細胞接着の誘導と抑制に関わる転写調節と翻訳後調節の機構

研究概要	リンパ球は炎症の際に病巣に遊走するだけでなく、平常時において特に病変のない場合にも日常的にリンパ節・皮膚・腸管などの組織にホーミングする。炎症反応の際も、平常時の組織へのホーミングにおいても、セレクチンとその糖鎖リガンドを介した細胞接着が関与する。炎症反応の際のリンパ球の炎症局所への動員と、平常時の諸組織へのホーミングとでは、関与するセレクチンのリガンドが異なる。硫酸基をもたない通常のシアリルLe^xは主として炎症時のリガンドであるのに対し、我々が最近同定したセレクチンの新規リガンドである硫酸化シアリルLe^x(シアリル6・スルホLe^x)は、平常時のホーミングで活躍するリガンドである。本研究においては、リンパ球のこの二種のセレクチンリガンドの機能分担と発現調節機構を、これらの糖鎖リガンドの合成酵素の転写調節のレベルと、我々が最近見出した翻訳後調節の機構の両面から解明を試みた。炎症反応の際には硫酸基をもたない通常のシアリルLe^xの発現が誘導されるが、この誘導は合成酵素フコシルトランスフェラーゼVIIの転写誘導による。今年度の検討により、この酵素遺伝子の転写誘導に、転写因子THP-1(Gli-2)の関与があることが判明した。また、硫酸化シアリルLe^xについては、合成酵素であるGlcNAcβ:6-スルホトランスフェラーゼについて検討し、リンパ球系細胞の平常時のホーミングで活躍する硫酸化糖鎖リガンドの合成には、GlcNAcβ:6-スルホトランスフェラーゼの五種のアイソザイムのうちで、GlcNAC6ST-1およびHEC-GlcNAC6STが関与することが明らかになった。このうち、GlcNAC6ST-1については、炎症性サイトカインであるIL-1βやTNF-αによって転写誘導が認められた。また、GlcNAcβ:6-スルホトランスフェラーゼの産物は、セレクチンを介した細胞接着のみならず、セレクチンと同じくリンパ球のホーミングに深く関与する接着分子CD44の細胞接着活性の調節においても重要な役割を演じていることが明らかになった。
