クロマチンリモデリング因子Mi-2による細胞増殖制御機構の解明

研究概要

ショウジョウバエのDNA複製/増殖関連遺伝子を共通に制御する転写因子DREF(709aa)が、クロマチンリモデリング複合体のサブユニットとして知られるMi-2(ATP依存性ヘリカーゼ)と相互作用することをショウジョウバエを用いた遺伝学的な方法や免疫沈降法などの生化学的方法により証明した。両者の結合領域をGST pull down方を用いて決定したところ、Mi-2はDREFのDNA結合ドメインに結合することが明らかとなった。さらにMi-2はDREFのDNA結合活性を阻害し,細胞内で相互作用した際にはDREFの転写因子としての機能を抑制的に制御していることを示唆した。さらに、GAL4-UAS系を用いたDREFとMi-2の過剰発現系統や、Mi-2の変異体を用いた遺伝学的な解析の結果、Mi-2の活性もDREFにより抑制的な調節を受けている可能性が示唆された。また、ショウジョウバエ唾腺染色体におけるDREFとMi-2の局在を免疫染色法で調べると両者は相互排除的に染色体に結合していることを観察している。これらの結果から、クロマチンリモデリング因子と転写因子との特異的な結合により相互の活性を負に調節するメカニズムがあるらしいことや、転写の調節にはクロマチンの構造変化の調節が重要な意味を持っていることを示唆することができた。
