哺乳動物上皮細胞の接着構造形成過程におけるnPKCηの役割の研究

研究概要	本研究は、プロテインキナーゼC(PKC)分子種の一種であるatypical PKC(aPKC)群が、「上皮細胞の接着構造形成に決定的な役割を果たすことを介してそのapico-basalな上皮細胞極性の発達に寄与している」という我々のこれまでの知見に端を発した研究課題である。すなわち、aPKCはTPA非感受性であるにも関わらず、「低カルシウム培地培養によって細胞間接着を解離させた上皮細胞にTPAを添加するとE-cadherinのcell-cellへの凝集を伴わないTJ形成が誘導される」という現象がなぜ起こるのかを明らかにしようとする研究である。この現象は、上皮細胞接着形成に他のTPA感受性PKCも関与している可能性を示唆していたが、実際にこれまでの研究を通じて、「上皮系の細胞に特異的に発現しているnPKCηが、E-cadherinからの初期接着刺激とaPKCの活性化をつなぐ位置で働いている」ことを示唆する結果を得ることができた(昨年度実績報告書参照)。この成果の上に立って本年度は、このnPKCηの働きを分子レベルで解明することを目的として研究を進めた(研究計画参照)が、残念ながらこの点においては十分な成果を得るに至らなかった。但し、今回初めて「低カルシウム条件でもTPAで刺激すると、伸展し接触を始めた細胞間にE-cadherinが初期に一時的に集積する」という事実を発見することに成功し、このことにより、先のnPKChの優勢抑制変異体の効果とあわせて、「nPKCηがE-cadherinの集積過程に働くことを介して細胞接着構造形成に寄与している」という仮説を立てることが可能になった。現在、こうした知見に基いて、本研究課題の成果を論文にまとめる作業を進めている。一方、aPKC-PAR systemの接着構造形成に果たす役割に関しては大きく研究が進み、裏面に記載した諸成果を本年度に公表することができている。
