研究概要	本研究では、分泌性ホスホリパーゼA2(sPLA2)によるアラキドン酸代謝のSwitch-onを指標に、細胞膜マイクロドメイン中のリン脂質が細胞膜活性化に呼応して如何なる修飾を受け、これがどのように細胞応答につながるのかについて、分子レベルで明らかにすることを目的として研究を進めた。すでに申請者はこれまでに、sPLA2群を(1)IIA型sPLA2タイプ:炎症性刺激により活性化状態にある細胞膜に作用する、(2)X型sPLA2タイプ:未刺激の細胞膜に作用する、(3)V型sPLA2タイプ:(1)と(2)の混合タイプ、に分類できることを報告してきた。本研究では、新規sPLA2であるIID, IIE, IIF, III型sPLA2について(1)〜(3)のタイピングを行い、IIDとIIEは(1)、IIFとIIIは(3)に属することを明らかにした。(1)に属する酵素はホスファチジルコリン(PC)に対する酵素活性が極めて弱いためPCに富む形質膜外層には作用しないが、ヘパラン硫酸プロテオグリカン(HSPG)への結合を介してカベオラ/ラフトに集積し、刺激依存的に細胞内小胞に取り込まれ、核周縁に集積して機能する。(2)の酵素はHSPGへの親和性を示さないが、PCに対する活性が極めて高いため、形質膜外層のPCを容易に加水分解できる。(3)の酵素はPCに対して有意な活性を示すため(2)と同様に形質膜外層に作用できるが、HSPGへの親和性も有するため、(1)の経路も利用できる。本研究を通じて、sPLAの酵素的性質・局在性と膜のダイナミクスが適切に連動した時に初めてsPLA2がアラキドン酸遊離機能を発揮できることが明らかとなった。
