後部帯状回における扁桃体および海馬体機能の統合による行動出力機構

研究概要

大脳新皮質の内側にある大脳辺縁系は、扁桃体、海馬体、帯状回などの領域を含み、情動、空間認知、エピソード記憶などに重要な役割を果たしている。本研究では、覚醒行動下ラット後部帯状回から同時に複数の単一ニューロン活動を記録し、空間的な感覚刺激-報酬連合学習課題に対する応答性を調べ、辺縁系における情報処理の統合機構の解明を試みた。
平成14年度は、1)ニューロン活動の記録を遂行し、完了させ(担当:田淵)、2)記録した複数のニューロン活動をクラスター解析により単一ニューロンに単離し(担当:堀・田淵)、3)対照期、感覚条件刺激呈示期、遅延期および報酬獲得期(リック行動期)に分けて後部帯状回ニューロンの応答性を調べ(担当:田淵・堀)、4)さらに相互相関解析による後部帯状回ニューロン間のシナプス結合性について調べ(担当:田淵)、5)他の辺縁系部位のニューロン応答性と比較・解析して行動発現における辺縁系内機能局在の解明を試みた(担当:田淵)。
これらの結果、1)多くの後部帯状回ニューロンが脳内自己刺激や蔗糖溶液を獲得する報酬期に応答した。2)報酬と連合したにもかわらず感覚条件刺激呈示期にはほとんどのニューロンが応答しない。3)後部帯状回にはリック行動と非常に高い相関を示すニューロンが存在する。4)とくに3)のニューロンはリック行動数十ミリ秒後に活動がみられたことから運動のモニターに関連していることが新知見としてみつかった。
以上より、後部帯状回は海馬体や扁桃体から報酬および運動情報の入力を受け、運動の評価を行っていることが示唆された。
