大脳皮質-視床下核-淡蒼球路の機能を探る

研究概要	大脳皮質の広い領野に由来する情報は、大脳基底核で処理された後、大部分は視床を介して大脳皮質、とくに前頭葉に戻るというループ回路を形成している。線条体ニューロンが淡蒼球内節に単シナプス性に投射する経路(直接路)と線条体ニューロンが淡蒼球外節と視床下核を経由して多シナプス性に淡蒼球内節に至る経路(間接路)の2経路を介して、大脳基底核の入力部から出力部に情報が伝達されるとされているが、それ以外にも大脳皮質-視床下核-淡蒼球内節という経路(ハイパー直接路)も存在する。実際、大脳皮質を電気刺激し、淡蒼球内節から神経活動を記録すると、早い興奮・抑制・遅い興奮という、3相性の応答を示す。これまでの研究から、早い興奮はハイパー直接路を、抑制は直接路を、遅い興奮は間接路を経由していることがわかってきた。本年は、ハイパー直接路や間接路が実際、どのように働いているか明らかにするために、視床下核にムシモル(GABA_A受容体作動薬)を注入することによりブロックし、淡蒼球ニューロンの発射活動に対する影響を調べた。(1)視床下核をブロックすると淡蒼球ニューロンの平均発射頻度が減少するとともに、バースト状発射を示した。(2)感覚刺激に対する応答が、明確でなくなった。ニューロンによっては、視床下核ブロック前後で、反対の反応を示すものもあった。(3)このような変化は、淡蒼球内節よりも淡蒼球外節で、より明確に観察された。以上の結果は、視床下核-淡蒼球投射が、淡蒼球ニューロンの自発活動や、感覚応答の形成に重要な役割を果たしていることを示している。
