各種近交系由来ES細胞の樹立

研究概要	現在使用されているES細胞のほとんどは免疫学的、神経科学的あるいは発生学的なデータの少ない129系統マウス由来のものである。そのため、多くの場合、遺伝的背景がことなりあらゆる情報の整った系統マウスに永い歳月を費やしバッククロスを行っている。そこで、遺伝子改変動物を作出する時から、遺伝的は背景のしっかりした系統動物由来のES細胞を用いることができればあらゆる点で有利になる。 1 我々がすでに樹立したES細胞のうち、BALB/c系ES細胞(ST/c)について、その性状を確認した。その結果、長期間凍結保存しておいたが、正常に生殖系列系のキメラマウスを得ることができた。3匹の100%キメラマウス全てが生殖系列系であり、そのうちの1匹は100%生殖系列系にトランスミットしていた。この結果から129/SvJ由来のES細胞E14.1およびBALB/c由来ST/cESを用いてノックアウトマウスの作出を試みた。E14.1ESでは2つの陽性クローンのうち、1つのクローンのみ生殖系列系キメラマウスが得られた。 ST/cについては多数の陽性クローンが得られたので、現在キメラマウスの作出を試みているところである。 2 その他、C57BL/6J系ES(ST/b)についても129系統のESと比較しながら、ノックアウトマウスの作出を計画中である。 3 本年はNZB系統のES細胞の作出をこころみた。その結果、60個の8細胞期胚から、1つのES様細胞の樹立に成功した。現在この細胞が生殖系列系であるかどうかの確認を行っているところである。
