癌免疫治療薬への応用を目指す組換え型二重特異性抗体の蛋白質工学

研究概要

本研究は、当研究室により世界で初めて巻き戻しによる調製に成功し、抗腫瘍効果を実証してきた癌抗原MUC1とリンパ球抗原CD3を標的とした最小の二重特異性抗体(Mx3 diabody)のさらなる機能改変を目的としている。今年度は、(1)二種のdiabodyの作製と併用の効果、(2)低免疫原性のエンハンス分子との融合、(3)他の癌抗原抗体を用いた汎用性の拡大、の観点から研究を進めた。実績は以下の通りである。(1)我々がクローニングした、リンパ球に共刺激シグナルを伝達する抗CD28抗体を用いて新たに作製したMx28 diabodyは単独でも抗腫瘍効果が見られ、さらにMx3 diabodyとの併用試験において、各々を混合した時よりもより低濃度で同等の効果、即ち相乗効果を得た。(2)現在、エンハンス分子として用いている黄色ブドウ球菌由来の変異型スーパー抗原(mSEA)に比べ、より低免疫原性であるヒト由来のリンパ球共刺激分子4-1BBLの巻き戻しによる調製系を世界で初めて確立し、diabodyとの融合を行ったところ従来のmSEAとの融合分子と同等の効果を得た。また同様に初めて巻き戻しによる調製に成功した抗腫瘍サイトカイン、ヒトinterleukin-12と抗体可変領域(Fv)との融合蛋白質を構築した。機能評価した結果、融合により抗腫瘍効果の大幅な上昇を観察した。これは抗体との融合により、癌細胞近傍へ抗腫瘍サイトカインを効率よく輸送出来たためと考えられる。(3)より普遍性が高い癌胎児性抗原(CEA)に対する抗体T84.66を用いて作製したCx3 diabodyは、従来のdiabodyと同等の抗腫瘍効果が様々な癌腫に対して見られた。以上により、抗腫瘍効果を有し、低免疫原性かつ汎用性のある新規癌免疫治療薬の可能性を十分に示すことが出来た。
