遺伝子組み換えバベシア原虫の作製法の確立とその応用

研究概要

本研究の目的は、バベシア原虫体内に異種の標的遺伝子を含む発現カセットを導入し、それらを原虫遺伝子内に組み込ませることで目的の蛋白質を恒常的に発現しうる遺伝子組み換え原虫の作製法を確立することにあった。
まず、真核細胞用CAG promoter、Toxoplasma gondiiのGRA1 promoter、もしくはBabesia bovisのRAP-1 promoterを上流に添えたenhanced green fluorescent protein (EGFP:蛍光発色蛋白質)発現カセットを含む3種類のtransfer vectorを構築し、エレクトロポレーション法によりBabesia bovis虫体内に遺伝子の導入を試みた。その結果、T. gondiiのGRA1 Promoter付きEGFP vectorを導入したバベシア原虫のみ、明瞭な蛍光色を細胞質内から発する陽性像を観察した。しかしながら、そのような陽性原虫の出現率は約0.01%と低く、また遺伝子導入後4日目にはその陽性蛍光像が完全に消失してしまったことから、更なる改良が必要とされた(Fujii et al.,J. Protozool. Res.投稿中)。次に、薬剤耐性遺伝子として知られるTgDHFR-Ts遺伝子を活用して遺伝子導入原虫を選抜し、陽性原虫の回収効率を向上させる試みを行った。そこではまず、TgDHFR-Ts蛋白質が耐性能を示す薬剤Pyrimethamineのバベシア原虫に対する増殖阻害効果が調べられた。その結果、IC_<50>、0.8-3.2μg/mlでPyrimethamineがパペシア原虫の試験管内増殖を有意に抑えることが示された(Nagai et al.,Antimocrob. Agents chemother.,2003)。そのため、Pyrimethamine耐性TgDHFR-Ts遺伝子及びEGFP遺伝子を共発現できる新たなTransfer vectorを構築し、Pyrimethamine選択下でのEGFP遺伝子発現B. bovis原虫の出現率及びその発現性状を現在検討している。
以上の成果から、1)エレクトロポレーション法によりTransfer vectorを原虫内に導入できること、2)T. gondiiのGRA1 promoterによって外来遺伝子をバベシア原虫体内で発現させることができること、3)一過性ではあるが、外来遺伝子EGFPをバベシア原虫体内で発現できること、4)Pyrimethamineがバベシア原虫の増殖抑制効果を示すこと、が明らかとなった。TgDHFR-Ts遺伝子を用いた遺伝子導入バベシア原虫の回収効率の向上に向けた研究は現在もなお進行中である。
