T細胞増殖応答におけるARNTの生理機能

研究概要	転写因子ARNTはbHLH-PASファミリーに属しており、同じファミリーのArylhydrocarbon Rceptor(AhR)やHypoxia Inducible Factor-1α(HIF-1α)とヘテロ2量体を形成することにより作用発現し、薬物代謝あるいはエネルギー代謝への関与が報告されている。本転写因子は胸腺にも高発現が認められているがその具体的な機能については明らかにされていない。また、代表的なダイオキシンである2,3,7,8-tetrachlorodibenzo-P-dioxin(TCDD)による胸腺萎縮とそれに伴う免疫抑制についてAh Rの関与が報告されているが、この毒性発現メカニズムについて不明な点は多い。今回我々はARNTのvivoレベルでの生理機能の解析と上記現象の分子メカニズムをより明確にするために胸腺細胞特異的ARNT欠失マウスを作成した。 ARNTに対する従来のジーンターゲッティング法では胎生致死(胎生9.5〜10.5日)であることが既に報告されていたので、Cre/LoxPシステムによりARNT遺伝子のDNA結合ドメイン(bHLH)を胸腺細胞特異的に欠失させた。得られた胸腺細胞ではARNT蛋白質は完全に消失しており、このシステムによる変異ARNT蛋白質によるdominant negative効果はないものと考えられた。また、それらのホモマウスは見掛け上正常に生まれ、T細胞系組織における分化マーカーは対照群と比較して変化はなかった。しかし興味深いことに、50μg/kgのTCDD投与実験において、投与後3日目の胸腺は、ヘテロマウスでは野生型と同様に、約4分の1の大きさに退縮するが、ホモマウスでは退縮は全く観られず、細胞数及びCD4/CD8の発現パターンにも変化はなかった。これらの結果より、ダイオキシン類による胸腺萎縮に、胸腺細胞に発現するARNTが関与することが明らかになった。今後、本遺伝子導入マウスを利用し、ダイオキシンを含む、多環系芳香族分子による、胸腺退縮機構を明らかにしていきたい。
