組み換え単純ヘルペスウイルス(HSV)による脊髄炎の発生病理

研究概要

背景
HSVの感染はウイルスの膜蛋白と細胞表面蛋白との結合、融合を経てウイルスゲノムが細胞内に侵入する事で成立する。我々はこの過程においてgDに注目し、我々の保有する様々な毒性を示すHSV2株について解析を試みた。
方法
(1)HSV2の沖縄株(K1〜K4)とチェンマイ株(C1〜C4)のgD領域の塩基配列の比較・検討
沖縄株4株とチェンマイ株4株をそれぞれVero細胞に感染させcytopathic effect(CPE)が生じた時点で超遠心法でウイルスDNAを抽出する。Bgl IIで切断した後L fragmentを回収し、pBluescript IIにクローニングする。塩基配列を決定し、株間での塩基配列の相同性や繰り返し配列の出現頻度さらにGC含量を比較する。
(2)HSV2のgD欠損株の作製
(a)recombinant plasmidの作製
(b)recombinant virusの作製
(c)gD rescue株の作製
(3)マウスヘのgD欠損株あるいはgD rescue株の接種
マウスにgD欠損株やg Drescue株を接種し、野生型との毒性の違いを検討する。
(a)病変の拡がりや程度を組織学的に検討する。特に病変部位におけるウイルスの存在をin situ hybridizationで確認する。
(b)ウイルスの感染臓器をPCRで検出する。
結果
チェンマイ株(C1〜C4)は沖縄株(K1,K3とK4)より毒性が強い事を報告したが、これらをVero細胞に感染させ、CPEの出現を比較したところ、時間的な差は軽度あるもののどの株も全ての細胞にCPEが現れた。従って、in vitroでは毒性の違いは軽度であるが、ウイルスを投与した場合は宿主の免疫系の違いや神経細胞に対する親和性の違いなどにより病原性の差が明確になる事が分かった。
