腸管出血性大腸菌の持つ様々な病原因子遺伝子の多様性に関する研究

研究概要	腸管出血性大腸菌は多くの病原性因子を持つことが知られており、個々の因子に関する研究は進みつつある。しかし、病原因子遺伝子の多くには多様性(変異)があり、この多様性がどの程度存在するのか、また、多様性がどのようにヒトへの病原性に影響を与えるかについてはあまり調べられていない。本研究は腸管出血性大腸菌の持ついくつかの病原因子の多様性を調べることにより、それぞれの因子の機能解析を行うことを目的とする。 Stx遺伝子についてはほぼ解析を終えているため、次にrpoSの解析を行うこととした。rpoSは腸管出血性大腸菌の持つストレス耐性(酸、凍結、乾燥など)に関与しており、ストレスに応じる遺伝子の発現制御を行っていると考えられている。ABI Genetic Analyzer 310を用いたCapillary Electrophoresis SSCP(CE-SSCP)法を用いて分析を行い、臨床分離株におけるrpoSの変異は5'側から中間領域にかけて存在し、3'側は保存されている。これらの株における多様なSSCPパターンについてDNA塩基配列を決定することにより、変異とSSCPパターンとが相関していることを確認した。現在、これらの変異が腸管出血性大腸菌の他の病原因子の発現に及ぼす影響について解析している。
