研究概要	悪性黒色腫(メラノーマ)は神経堤細胞に起源をもつ色素細胞(メラノサイト)から発生し、、所属リンパ節のみならず肺や皮膚に遠隔転移をきたす最も予後の悪い悪性腫瘍である。平成13年度においては、運動能と浸潤能がすでに評価された7種類のメラノーマ細胞株(低運動性:3種類、高運動性:4種類)、及び正常皮膚メラノサイトを用い、Affymetrix社製GeneChip^<TM>によって約6800個の遺伝子について各々の遺伝子発現の差異を解析し、細胞の運動性に共通した遺伝子発現のパターンの解析を行った。Silicon Genetics社、GeneSpring^<TM>を使用し、階層分析を行った結果、遺伝子発現のパターンからメラノーマ細胞株を低運動性と高運動性に分類することができた。また、細胞外マトリックスに対する接着分子・糖蛋白質、あるいは蛋白分解酵素の活性化にかかわる遣伝子にも共通した発現パターン結果が得られた。特に浸潤性の高い細胞では細胞周期関連タンパク質の発現とよく相関することが判明し、従来まで個別に捉えられてきた細胞増殖機構と細胞運動能について改めて検討すべき課題であろう。一方で、免疫担当細胞に発現するCCR7、CCR10、CXCR4などのケモカイン受容体が通常発現していないメラノーマ細胞に見られたことは、メラノーマ細胞が固有の臓器親和性をもって転移する機構が、免疫担当細胞が個々の臓器に親和性をもって遊走する機構と共通することを示唆している。
