ヒト正常造血前駆細胞のSrcファミリーキナーゼによるアポトーシス制御機構の解明

研究概要

ヒトの造血系ではFas/Fas ligand(Fas-L)システムやtumor necrosis factor α(TNF-α)などによるdeath domainを介したアポトーシスが造血調節に深く関与することが知られており、この機構の破綻が各種の疾患を引き起こすと考えられる。一方、stem cell factor(SCF)やerythropoietin(EPO)などの造血因子は血球の分化・増殖に働く。また、その存在そのものがアポトーシス回避に働くことが報告されているが、これらの生存シグナルがdeath domainを介した死のシグナルにどのように拮抗するか、その詳細は不明である。SCFやEPOにはSrc family kinaseを介し、生存を維持する働きが想定されている。我々はSrc family kinaseの一つ、Lynに注目し、Fas/Faas-L系を介したアポトーシス回避への関与を報告した。(Exp Hematol 29 : 19, 2001)また、Src family kinaseの下流にあるAKTがSCFを介したアポトーシス抑制において重要な役割を果たしていることも報告した。(Exp Hematol 29 : 838, 2001)これらの基礎的な事象は現時点ではすぐに臨床応用可能なレベルへは至っていない。しかし、種々の造血幹細胞移植を行い、かつその有用性を報告してきた我々にとって、それによっても治癒しない症例も存在していることへの回答としてこれら分子病態的なアプローチの必要性が示唆された、と考える。今後は臨床応用を念頭においたドラッグデザインなどへの発展を予定している。また、他のアプローチとしてヒト赤芽球系前駆細胞に発現する転写因子ZNF317の新規クローニングに成功し、その機能解析を実行した。
