DNAマイクロアレイ法を用いたFKHRL1のターゲット遺伝子の同定と機能解析

研究概要	FKHRL1はForkhead転写因子ファミリーに属する転写因子である。同ファミリーはForkhead domainと呼ばれるDNA結合ドメインを有し、線虫からヒトまで、広く保存されており、胚形成、分化、腫瘍化などの機能を担っていることが推察される。その一員であるFKHRL1は二次性白血病で認められたt(6;11)(q21;q23)転座におけるMLL遺伝子の新しい融合相手として同定され、線虫の寿命に関連しているDaf-16のヒトホモログである。我々は、このFKHRL1が、エリスロポエチン(EPO)シグナル伝達において主要な伝達経路の一つであるP13-キナーゼ-Aktの下流に存在し、活性化されたAktによって直接リン酸化されることを報告した。また、臍帯皿をCD34+、CD34+/38-、CD41+、GlycophorinA+分画に分け、半定量的PCR法を用いてFKHRL1のmRNA発現量を比較し、GlycophorinA+の分画に多いことを確認した。以上より、FKHRL1は正常赤芽球系造血において何らの役割を担っている可能性が示唆された。しかし、FKHRL1の標的分子は同定されておらず、その機能も不明である。FKHRL1は非リン酸化状態で核内に存在し転写因子として働くが、リン酸化を受けると核外に移行し転写活性を失うことが知られている。我々は、Aktによってリン酸化される3つのアミノ酸残基をアラニンに変換した恒常活性型変異体(FKHRL1-TM)をEPO依存性細胞株UT-7/EPOにタモキシフェン誘導系を用いて遺伝子導入し、転写活性を持つFKHRL1の発現を誘導操作しうる系を確立した。今後は、この細胞からタモキシフェン添加の前後でRNAを抽出し、DNAマイクロアレイ法を用いて発現量に差のある遺伝子を見つけ、real time PCRで定量化し、FKHRL1の標的分子の同定を試みる予定である。
