14-3-3σによる上皮細胞の最終分化、G2/M期調節機構の解明

研究概要

重層扁平上皮を構成するケラチノサイトを用いて、上皮特異的プロテインキナーゼC:PKCηとその結合蛋白質14-3-3σの増殖阻害、分化誘導作用機構の解明を進めた。
PKCηは、CDK2/CyclinE/p21と相互作用し、G1期停止を誘導する。
1)PKCη/CDK2/CyclinE複合体のアダプター蛋白質として、14-3-3ζ及び14-3-3σを同定した。14-3-3ζが、他のPKC分子種(PKCα,PKCδ,PKCζ)とも相互作用するのに対して、14-3-3σはPKCηにのみ結合した。またその結合部位は、PKCηC末端領域であった。
2)14-3-3σは、PKCηとの共発現により増殖停止、分化誘導を相乗的に促進した。
3)14-3-3σは上皮組織に限局して発現する。表皮では有棘層、顆粒層に存在し、これはPKCηの発現部位と一致する。細胞内ではPKCηと同様、核周辺部に存在した。しかし、ケラチノサイトの分化に伴い、14-3-3σは核へと移行した。この局在変化は、14-3-3のリン酸化レベルに依存していることが示された。
4)14-3-3σの核移行に伴い、細胞は老化様形態を示し、老化マーカー:senescence assooiated β-galactosidase活性(SA-β-gal)が誘導された。しかしながら、p16INK4,p19ARF,p21WAF1/Cip1の発現には大きな変化が認められず、テロメア長の変化も見いだされなかった。従って、これ以外の老化メカニズムが関与していることが予想される。
