心筋への遺伝子導入による新しい心不全治療法の開発

研究概要

(1)SR Ca2+-ATPase遺伝子の心筋内遺伝子導入による新しい心不全治療法の開発
human SR Ca2+-ATPase遺伝子をアデノ随伴ウイルスベクターおよび、レンチウイルスベクターに組み込み、遺伝子導入ベクターを作製した。培養心筋細胞に遺伝子導入したところ、ふたつのベクターともにSR Ca2+-ATPase mRNA発現量は約3倍に増加し、また、SR Ca2+-ATPase蛋白も約2倍にまで増加した。Ca2+輸送能の亢進が認められ、治療効果が確認された。現在、動物実験モデル(心不全モデル)で効果を検討中である。
(2)phospholamban遺伝子翻訳の特異的抑制による心不全治療法の開発
phospholambanの機能を抑制するため、human phospholamban遺伝子のmRNA発現をRNA interference法によって遺伝子特異的に抑制することに成功した。human phospholamban遺伝子の蛋白翻訳領域の21塩基に相同な二本鎖RNAを合成し、HVJ-envelopeを用い培養心筋細胞に導入した。発現に対する効果は、きわめて遺伝子特異的であり、PLB mRNA発現を対照群の約10%にまで低下させるものの、同じ心筋小胞体蛋白であるSR Ca2+-ATPaseやcalsequestrin mRNAに対しては全く抑制効果を示さなかった。また、この遺伝子レベルでの変化は蛋白レベルにも反映され、phospholamban蛋白は対照群の10%以下に抑制され、その効果は1週間持続した。さらに、心筋小胞体へのCa2+汲み上げ能はこの方法により38%亢進が見られた。
