研究概要

病態毎に多数の症例を混合してSAGEライブラリーを作製し、各病態を代表すると思われるプロファイルの解析を行った。これによって作製された肝臓由来のライブラリー数は10個(約50万発現遺伝子tag)となり、正常肝臓、肝炎ウイルス感染肝臓、肝細胞がんにおける発現遺伝子ライブラリーが得られた。これによって現時点では世界最大の肝臓SAGE情報となった。肝細胞がんの発現遺伝子プロファイルは、正常肝組織、背景の肝硬変組織と異なることに加えて、感染している肝炎ウイルスの種類によっても異なることが明らかになった。同時に、肝細胞がんであっても、多くの肝臓特異的遺伝子発現が維持されていることも示された。この結果は、肝硬変結節から、前がん病変、さらには肝細胞がんへの発現遺伝子の変化をとらえるには、詳細な遺伝子解析を行う必要があることを示唆していた。各SAGEライブラリーには、ESTにも登録されていない発現遺伝子tagが数多く存在し、肝細胞がんに高い発現を認めるもの、あるいは発現が低下しているものが多数見いだされた。これらがヒトゲノム上に存在することを確認し、特徴的な112個についてはゲノム情報をもとにプローブを作製し、これまでに13個についてはノザンハイブリダイゼーシヨンにても特異的な発現を確認している。こうした遺伝子の構造と機能を明らかにすることは肝がん研究のみならず、がん研究に極めて重要であると考えている。
これまで得られたSAGE情報とDNAチップ解析の結果をもとに、肝硬変および肝細胞がんの発現プロファィルを特徴づける遺伝子を選択・収集した。これに加え、先のSAGEライブラリーで2tags以上の発現し、肝臓で特異的に発現している遺伝子を収集し、あわせて約12,000個の肝臓において発現している遺伝子を保有した。
