棘皮動物卵を用いた受精の神経生物学的解析

研究概要	卵・精子の活性化等については従来主に生理化学的な手法で研究されており、活性化に必要な細胞内情報伝達系をイオンチャネルがトリガーすることが示唆されてきた。すなわち、このような非神経細胞においてもイオンチャネルが細胞外の環境変化に対するセンサーとして働き、細胞内の情報伝達系を活性化しているのである。我々はそのメカニズムをより直接的に探るため、従来この分野ではあまり用いられてこなかった電気生理学や分子生物学の手法を用いた研究を計画した。ウニ卵は大型球形のため微小電極を用いた細胞内記録,2電極膜電位固定,パッチクランプなど各種電気生理学的手法を適用しやすい。この利点を活かして,まずバフンウニ、タコノマクラ、アカウニ、イトマキヒトデ、ユウレイボヤの卵を用い受精電位および活動電位の測定を行い、受精電位をトリガーする精子・卵相互作用の情報伝達系路と電位生成に関与するイオンチャネルメカニズムの解析の基礎的なデータを得た。また、未受精卵に存在するイオンチャネルをパッチクランプおよび2電極膜電位固定法を用いて解析した。それらの結果、カルシウムチャネルおよび少なくとも3種類のカリウムチャネル(内向き整流性、遅延整流性、およびいわゆるA電流型の電流が同定できた。さらに、表層穎粒開口放出のメカニズムを解析するための基礎として、蛍光色素FM1-43をウニ卵にロードして受精の前後における蛍光強度の変化をCCDカメラを用いて計測し、イメージングソフトウェアを用いて解析した。
