植物根-土壌界面における分泌現象の実態解明

研究概要	通常、植物の根の表面は粘液等の分泌物に覆われている。この粘液層は水や無機塩の植物体への取り入れを介在する働きをもつだけでなく、土壌のさまざまな物理・化学的ストレスから根を保護することが示唆されている。粘液層は以上のような重要な働きを担うにもかかわらず、この層がどのような土壌条件においてどの程度根の表面を覆っているのかは全く明らかにされてはいない。本研究では、植物根と土壌界面における分泌物の拡散範囲とその消長を総合的に把握することを目的とする。本年度は、まず植物根の根圏土壌中での粘液分泌動態を視覚的に明らかにすることと、根圏土壌内の安定同位体炭素および重水の濃度勾配から、分泌物の土壌中への拡散範囲を定量的に把握する方法論を検討した。根は微弱な光刺激により生育阻害を起こすため、高解像度暗視野カメラを使用し、透明なアクリルパネル越しに土壌中を伸長する植物根の表層部を観察するシステムを組んだ。粘液物質の動態把握方法を検討し、画面上の粘液で覆われた面積と実際の粘液の大きさとの相関関係を求めたところ両者には高い正の相関関係を認めた。次に、トウモロコシを^<13>CO_2とD_2Oで暴露し、根圏土壌中に放出される安定同位体の存在量を分析し、13CおよびD_2Oの根圏内濃度勾配を検出し得るような方法論を検討した。同位体炭素については土壌を直接燃焼させることにより、また重水については遠心分離法により得られる土壌水を集め、既設の安定同位体比質量分析計およびH-Deviceにより測定した。その結果、根に付着した土壌鞘内の炭素と水の放出は確認できた。しかし、土壌鞘の外側への同位体の放出については、まだ充分には確認できていない。次年度に土壌鞘の外側への粘液拡散を捉え得るような実験系を検討する予定である。
