研究概要

[目的]:一昨年、成長ホルモン(GH)分泌刺激物質受容体の内在性リガンドとして、ラットの胃から28アミノ酸残基からなる新たなペプチド、ghrelinが単離された。我々はこのghrelinが摂食機構、GH分泌および胃酸分泌に関与していることを先に報告した(Nature 409:194-,2001;Biochem. Biophys. Res. Commun275:477-,2000)。今回、ghrelinが生後どの時期から胃に発現するのか?また新生児におけるghrelinの生理的役割を検討した。
[方法・結果]:胎児18? 20日齢および新生児の胃を摘出し、グレリンN端部に対する自家抗体を用いて免疫染色を行い、ghrelin陽性細胞を検索した。その結果、胎児18日齢において極めて小数であるがghrelin細胞を認めた。その数は生後、週齢が進むにつれて著しく増加し、広範囲に認められるようになった。次に、1、3週齢の新生児にghrelinの単一皮下投与を行い、15分後の血中GHを測定した結果、有意な分泌促進が認められた。さらに新生児に毎日ghrelinを皮下投与した結果、ghrelinは開眼あるいは膣開口までの日数の短縮をもたらした。
[考察]:ghrelinの生産が妊娠末期の胎児の胃で認められたことは、ghrelinが胎児の成長に関係している可能性を示唆している。新生児では、ミルクの制限によりghrelin分泌量が変化することを先に報告したが、今回の新生児におけるGH分泌促進効果や、膣開口日の短縮などは、新生児の胃の ghrelinがミルクの摂取の他に、成長や思春期到来においても関与している可能性を示唆している。
